INTERMEDIATES IN FLAVOPROTEIN CATALYZED HYDROXYLATIONS Barrie Entsch, Vincent Massey and David P. Ballou Department of Biological Chemistry, The University of Michigan Ann Arbor, Michigan 48104 Received November 21,1973 Summary The reaction of molecular oxygen with the complex of reduced p-hydroxybenzoate hydroxylase and 2,4-dihydroxybenzoate has been followed by rapid reaction techniques. During the reaction, which produces stoichiometric amounts of oxidized enzyme and the hydroxylated product,2,3,4-trihydroxybenzoate, three spectroscopically distinguishable intermediates have been detected and characterized.

Stepwise oxygenations of toluene and 4-nitrotoluene by a fungal peroxygenase.

Fungal peroxygenases have recently been shown to catalyze remarkable oxidation reactions. The present study addresses the mechanism of benzylic oxygenations catalyzed by the extracellular peroxygenase of the agaric basidiomycete Agrocybe aegerita. The peroxygenase oxidized toluene and 4-nitrotoluene via the corresponding alcohols and aldehydes to give benzoic acids. The reactions proceeded step...

Hidden intermediates and levinthal paradox in the folding of small proteins.

It has long been suggested that existence of partially folded intermediates may be essential for proteins to fold in a biologically meaningful time scale. Although partially folded intermediates have been commonly observed in larger proteins, they are generally not detectable in the kinetic folding of smaller proteins (approximately 100 amino acids or less). Recent native-state hydrogen exchang...

CueO is a multi-copper oxidase that confers copper tolerance in Escherichia coli.

The putative multi-copper oxidase CueO had previously been implicated in intrinsic copper resistance in Escherichia coli. In this report we showed that the presence of CueO in the periplasm protected alkaline phosphatase from copper-induced damage. CueO contained four copper atoms per molecule and displayed spectroscopic properties typical of blue copper oxidases. CueO catalyzed the oxidation o...

Oxidative metabolism of xenobiotics during pregnancy: significance of microsomal flavin-containing monooxygenase.

Pregnancy related changes in oxidative metabolism of model substrates were examined in CD1 mice. As compared to nonpregnant females, a significant decrease in the hepatic microsomal aminopyrine-but not in dimethylaniline-N-demethylase activity was observed in pregnant mice. The rates of microsomal flavin-containing monooxygenase-catalyzed N-oxidation of dimethylaniline remained relatively uncha...

Saturation behavior of superoxide dismutation catalyzed by the iron containing superoxide dismutase of E. coli B.

The iron containing superoxide dismutase from E. coli B is shown to catalyze superoxide dismutation by a mechanism which exhibits saturation kinetics. This behavior is quite different from that observed previously with bovine Zn/Cuand iron-containing superoxide dismutase from P. leiosnathi. Two parameters of catalysis were measured in the pH range 7.2 to 10.4: k was found to be independent of p...
